What is FOXO4?
FOXO4 (FOXO4-DRI) is a D-amino acid retro-inverso peptide designed to selectively target and eliminate senescent cells — aged, dysfunctional cells that accumulate with age and drive chronic inflammation and tissue dysfunction. It is one of the most targeted senolytic compounds available for research, acting through a specific protein-protein interaction rather than broad cytotoxicity.

Mechanism of Action
Senescent cells survive by upregulating FOXO4, which interacts with p53 to suppress apoptosis (programmed cell death). FOXO4-DRI disrupts this FOXO4/p53 interaction, restoring p53's ability to trigger apoptosis specifically in senescent cells. Because non-senescent cells do not rely on this FOXO4/p53 survival mechanism, FOXO4-DRI selectively eliminates senescent cells while leaving healthy cells intact. This targeted senolysis reduces the senescence-associated secretory phenotype (SASP) — the pro-inflammatory cytokine release that drives inflammaging.
